In vitro Propagation of Hyacinthus orientalis L.



Abstract
The study was carried out to develop year-round propagation system of bulb by redifferentiation of bulblet from bulblet scale or leaf tissue of Hyacinthus orientalis bulblet obtained through flower stalk tissue culture.
The experiments were conducted on totipotency of bulblet scale tissues in different ages and duration of low temperature storage at 4¡É and totipotency of bulblet scale tissues and leaf tissues cultured in different sucrose concentrations and pH values.
The results obtained were summarized as follows.
1. The 76days old bublet was identified as the most optimal age for bulblet formation and propagation of bulblets, and totipotency was better in bulb scale tissue excised from bulblets cultu red under light than under dark.
2. Percentage of organogenesis from bulblet scale tissue treated with low temperature for 9 weeks was slightly higher compared with other treatments, but proliferation of bulblets was rather better in bulb scale tissue without chilling treatment. medium containing 1.0§·/§¤ IBA was more effective for culture of both tissues than 1.0§·/§¤ IAA added one.
3. Leaf tissue was determined as better culture material compared with bulblet scale tissue, and its optimal sucrose concentration was 5.0%.
4. Percentage of plantlet differentiation from leaf tissue was also higher than from bulblet scale tissue, and its percentage of organogenesis was 100%, regardless of pH value. Optimal pH value for propagation was 5.8, but for growth of bulblet after differentiation was 4.8 or 5.3.
Considering the results, we found that culturing leaf tissue excised from bulblet, by 0.5§¯ in length in Murashige and Skoog¢¥s medium containing 1.0§·/§¤ IBA, 50g/§¤ sucrose and 8g/§¤ agar and adjusted pH 4.8 or 5.3 and growing bulblet removed leaf tissue in green house, Year-round propagation of bulb of Hyacinthus orientalis will be possible.
